The Copa America host nation came into the game as favorites, even with Neymar’s absence, as Bolivia had lost their last three games.
Philippe Coutinho scored twice after the break, with a penalty and header, to give his team a two-goal lead.
Everton added a stunning third, with five minutes left in the match.
Peru play Venezuela in Rio de Janeiro on Saturday in the second Group A game.
Brazil’s next match is against Venezuela in Salvador next Tuesday.
All 10 South American teams are participating this year, along with guest nations Qatar and Japan.
